Ntamack picked up the injury in France’s warm-up 30-27 win over Scotland on Saturday.
The 24-year-old limped off the field in visible pain and a later scan revealed the seriousness of the injury.
Matthieu Jalibert will likely replace Ntamach as starting fly-half.
France begin their World Cup journey on September 8 against New Zealand.
“It is absolutely devastating, especially when you are one of the home players, you have the opportunity to play at home and you are one of the favorites to win it,” former England international Ugo Moyne said.
The news is a huge blow for France as Ntamack is regarded as one the stars of the team with the 24-year-old also starring at the last World Cup for Les Blues in 2019.
