The veteran winger sustained the problem during non-contact training, his Premier League side announced.
The Croatian international will have surgery on his right knee before beginning his rehabilitation.
Ivan inked with the North London team on a 2-year deal from Inter in 2022, and he will be a free agent next summer.
His side has been unbeaten in 5 Premier League games under new head coach Ange Postecoglou from the beginning of the campaign, triumphing in 4 and drawing in 1, and Spurs are currently in second place in the league.
Perisic, who won the Champions League with Bayern Munich in 2019-20 and triumphed in Serie A with Inter in 2020-21, played 5 matches so far this season and came from the bench in all of them.
