The 33-year-old retired from the sport in 2020, but returned earlier this year after giving birth to two children.
She took part in the US Open in September, crashing out in round four to the eventual champion Coco Gauff.
Wozniacki clinched her only Grand Slam title in Australia in 2018 after a memorable victory over Simona Halep.
“I have so many wonderful memories of Melbourne, and of course winning the Australian Open is an all-time career highlight,” she said.
The tournament starts on January 14 and it will be extended by one day for the first time in history in order to cut down late-night finishes.
